Character design should reflect the characters' attitudes, personas, and quirks. Everything from the big things like their choice of clothes, their key facial expressions, the shape and sizes of their form... to what lines and angles are used in the style, what colors they wear, to their relationship in size and shape to their supporting characters. It's an art that few don't really grasp, but it's pretty subliminal.
